Leading to the August 2009 juvenile cancer warning, the FDA studied children and adolescent cancer patients who had been treated with TNF blockers. Federal drug safety officials identified an increased risk of cancer, occurring on average after 30 months of treatment. About half of the cancers were lymphomas, a type of cancer involving cells of the immune system. Some of the cancers in the FDA study were fatal.

Many patients with autoimmune diseases such as rheumatoid arthritis have used Enbrel (etanercept). This drug was approved by the U.S. Food and Drug Administration in 1998. Shortly after FDA approval, reports about Enbrel noted nearly 30 serious infections and six fatalities. This prompted an alert to doctors and required the manufacturer to expand the drug's warning label.

Since the approval of Enbrel the following serious conditions have been associated with some patients:

• Central nervous system disorders, including Multiple Sclerosis;
• Serious infections, including Sepsis;
• Tuberculosis;
• Blood disorders;
• Vision damage.
